00:00:10
so okay apart from uh genetic methods of
00:00:14
looking at this Gene gene therapy uh how
00:00:17
else what kind of can we do
00:00:20
now yes so we can also use chemical
00:00:25
activators because there is one way just
00:00:27
let's put more protein in mhm another
00:00:31
strategy well let's use the protein that
00:00:34
we already have that our cells are
00:00:36
already making and make it more active
00:00:40
because C six is an enzyme so we can
00:00:43
activate it enzymatically we can
00:00:45
potentially also try to trick cells into
00:00:47
making more of it uh but that's kind of
00:00:50
a different approach or we could
00:00:52
increase the activity of the
00:00:55
enzyme uh we are also exploring it uh
00:00:59
and there are several companies that
00:01:01
were looking for chemical activators of
00:01:04
C six so here we had some Edge because C
00:01:10
six is an enzyme and it has two separate
00:01:13
enzymatic
00:01:14
activities one is called detil so it
00:01:18
removes acle group from other proteins
00:01:21
and another one is ribosylation it adds
00:01:24
a ribos group so there are two
00:01:27
activities uh both are important for
00:01:30
various cellular
00:01:31
functions um but what we showed it was I
00:01:35
don't remember if we discussed this
00:01:36
Study last time or not that in human
00:01:39
centenarians we found a variant of C six
00:01:44
uh that has ration activity enhanced but
00:01:48
not
00:01:49
just and from this we concluded that
00:01:52
this is the activity we really want to
00:01:54
go after because acation is important no
00:01:58
question about it and I I believe it's
00:02:00
particularly important early on uh
00:02:03
during development when you know genome
00:02:07
has to properly function to make a baby
00:02:11
correctly but during
00:02:15
our uh you know subsequent life adult
00:02:18
life um perhaps rosil is what becomes
00:02:22
more critical for
00:02:23
longevity uh but all of the initial
00:02:27
screens for C six activated were focused
00:02:30
on the
00:02:31
culation and we started looking for
00:02:34
activators of rosil so we are doing it
00:02:37
right now we're screening
00:02:40
mhm uh but from the activators that were
00:02:44
discovered
00:02:46
previously uh we just tested them
00:02:48
because they all were discovered as
00:02:49
distilation activators and we thought
00:02:53
okay let's check maybe some of them do
00:02:54
both maybe some of them activate both uh
00:02:58
and one of them did both because most of
00:03:02
them really did nothing to to ration
00:03:05
they were only targeting one activity
00:03:07
that's but one was activating both and
00:03:11
that's a interesting molecule um called
00:03:15
fuken uh that is derived from brown
00:03:19
seaweed uh which is very safe people eat
00:03:22
brown seaweed it's sort of a staple of
00:03:26
Asian Cuisine in Japan in South Korea
00:03:30
people consume a lot of brown seaweed
00:03:33
and soups and
00:03:36
other other
00:03:38
appetizers um and so we got kind of
00:03:41
excited because these are countries with
00:03:44
highest life
00:03:45
expectances and they eat a lot of this
00:03:49
um so we really started investigating
00:03:53
further the effects of fidance from
00:03:58
seed
00:04:00
so right so I understand that you that
00:04:04
not all the fiden works right it's quite
00:04:06
a complex molecule that can be like
00:04:08
different um that's correct so Fon is
00:04:12
kind of a family of
00:04:14
molecules and their backbones are
00:04:17
similar but they are not the same in
00:04:19
every algae species there are
00:04:23
differences in in the structure how
00:04:26
those sugar Moes are organized relative
00:04:29
to each other and we've been testing
00:04:32
various
00:04:33
isolates of Fons and not every not each
00:04:38
one activates 2 six we found maybe every
00:04:43
third does others they have no effect we
00:04:47
even had some that inhibited C oh so do
00:04:53
you know what the kind of mechan
00:04:56
mechanism of action is like why it's
00:04:58
different
00:05:00
well probably because they have
00:05:02
different structures right and we are
00:05:05
trying to figure out why um we don't
00:05:09
know we don't know this minimal
00:05:11
structural unit of fiden that's
00:05:14
important and we are working with sugar
00:05:16
chemists right now to identify the
00:05:19
structural unit and I hope that once we
00:05:23
succeed then we can make a better drug
00:05:26
out of this but right now uh we can just
00:05:30
see right okay um so I believe you tried
00:05:37
to Co in in mice as a as a c six
00:05:40
activator right because as I said like
00:05:42
the previous one was was genetic so yes
00:05:46
can can you talk a little bit about what
00:05:48
you saw with Fon in
00:05:50
mice yeah that's that's actually very
00:05:53
exciting that you know the results
00:05:55
exceeded our expectations we uh started
00:05:58
giving
00:06:00
to aged mice uh sort of
00:06:03
middleaged not very old but about one
00:06:07
and a half year of age and we just kept
00:06:10
them on this supplement and we had
00:06:12
control group of mice that didn't
00:06:14
receive
00:06:15
fuken uh and then we were measuring the
00:06:18
Frailty scores every month uh we
00:06:21
measured the inflammatory cyto kindes in
00:06:25
the blood um and the physical activity
00:06:30
like you know the walking speed and grip
00:06:33
strength and various parameters and
00:06:37
there was clear benefit uh for the mice
00:06:41
uh taking FID and it was very clear it
00:06:44
continued um and then I think an update
00:06:48
to our previous conversation my fiden
00:06:52
group actually survived longer so it
00:06:54
even had effect on
00:06:57
lifespan the what kind of dose were you
00:06:59
giving them was it something that a
00:07:01
person would be able to
00:07:04
mimic yes it wasn't too high well we
00:07:08
basically we started
00:07:10
from um sort of crude preparation of FID
00:07:16
and it wasn't Ultra Pure because um we
00:07:20
wanted it to be kind of similar to what
00:07:22
humans would take because you can make
00:07:24
Ultra Pure fuken but then the it would
00:07:27
be cost prohibitive so we took partially
00:07:30
purified kind of crude uh preparation
00:07:33
not not just ground up seaweed but you
00:07:36
know first purification steps and we
00:07:39
were giving it to mice U mixed with
00:07:42
their Chow diet so it wasn't a huge dose
00:07:46
I think for a human you know translating
00:07:51
the dose between Mouse and human is not
00:07:53
linear and it's kind of difficult to say
00:07:56
because we are much larger and we have
00:07:59
different surface to volume ratio um but
00:08:04
I would say approximately four grams of
00:08:06
fuken would be similar for for a person
00:08:11
so for do not age right do you do you
00:08:14
test
00:08:16
Eiden is is yes yes so we test uh we
00:08:20
test their samples to make sure that
00:08:22
they activate uh C six and then then
00:08:27
they call it C six activate
00:08:31
right so I saw in one of your papers
00:08:34
that you talked about the activation of
00:08:37
cerx requiring fatty acids or at least
00:08:40
being enhanced in the presence of fatty
00:08:43
acids I believe so first of all GNA is
00:08:46
that correct and would there be any
00:08:49
advantage would that talk talk about
00:08:51
when you should take um aert sex
00:08:55
activator to try and have like fatty
00:08:57
acids around at the same time
00:09:00
yeah you know it's an interesting
00:09:01
question we haven't tested it in mice
00:09:04
because yeah potentially we could try to
00:09:07
further um give them different diets
00:09:10
with the fatty acids um so fatty acids
00:09:15
they were shown to activate the
00:09:17
aculation activity uh we didn't um I
00:09:21
think we tested it with ration we didn't
00:09:24
see much of an effect although I don't
00:09:28
know how that would play out in
00:09:30
combination with fuken that's again
00:09:32
something to try uh so could be Tred
00:09:37
potentially you know it's really
00:09:39
interesting but um it seems that
00:09:44
various sort of treatments or diets um
00:09:49
that from um people believe to be
00:09:52
healthy and that there is evidence like
00:09:55
color restriction even not similar
00:09:59
uh intermittent
00:10:01
fasting many of them activate C two in
00:10:05
six of course the effects may be quite
00:10:09
mild um but we see very interesting
00:10:13
relationship that often well if we know
00:10:16
that these types of fatty acids are good
00:10:18
and like other studies showed they're
00:10:20
good and then oh we find they activate
00:10:22
cin six so it's C six really seems to be
00:10:25
this kind of Hub uh controlling many
00:10:29
processes related to aging and this is
00:10:31
why just healthy lifestyle is
00:10:35
good but obviously we want to have
00:10:37
stronger effects and this is what we get
00:10:39
with with fid and s36
00:10:44
activator so one of the things C6 does I
00:10:47
believe is that it well it's part of the
00:10:51
the fixing the transposons is helping
00:10:54
package the chromatin so so basically
00:10:56
helping with the epigenetics so I saw
00:10:59
you also talked it said it would be
00:11:01
interesting to kind of combine C six
00:11:03
with reprogramming with like yamanaka
00:11:06
Factor
00:11:07
reprogramming um could you talk a little
00:11:09
bit about that and well a is that
00:11:11
correct and B could you talk a little
00:11:13
bit about how that would work or or is
00:11:16
SE six working totally differently from
00:11:19
the the amakas in terms of the way that
00:11:22
it handles you know it is a difficult
00:11:25
question you're asking and we've been
00:11:27
thinking a lot about this
00:11:29
uh because
00:11:32
um C six is doing something similar in a
00:11:36
way that it rejuvenates the epig genome
00:11:38
so our epig genome gets disorganized as
00:11:42
we age and this may be the driver of
00:11:46
Aging Al together uh but how do we
00:11:50
rejuvenate it because what yamanaka
00:11:52
factors are doing they just take
00:11:55
everything and kind of completely redo
00:11:59
redo it and just bring us back to this
00:12:02
um embryonic form um and that is
00:12:06
dangerous intrinsically because when
00:12:08
cells lose their identity they can then
00:12:10
turn into cancerous cells and this is
00:12:14
why people working on reprogramming they
00:12:17
uh working to develop strategies where
00:12:20
you do it just a little bit you you
00:12:23
don't go all the way you don't turn our
00:12:25
cells into embrionic cells which would
00:12:27
be lethal but we just do it just one
00:12:32
step two steps but not all the way um
00:12:35
what C six is doing here is you know
00:12:38
similar in a way that it rejuvenates but
00:12:41
it really doesn't change the fate of the
00:12:44
cell it doesn't even it doesn't even try
00:12:47
to do it so there is no danger of losing
00:12:49
cell identity um because with respect to
00:12:53
transposons C six just goes in and
00:12:58
recognizes transposons we don't
00:13:00
completely understand how and condenses
00:13:03
them into heterochromatin it doesn't try
00:13:06
to rewire the whole
00:13:08
system um so in this way yanaka factors
00:13:13
um work you know in a very dramatic way
00:13:17
they just change everything um C six
00:13:21
just comes in and there are some places
00:13:23
that losen up and it
00:13:27
tightens right so it's working in a
00:13:29
totally different way um and okay so
00:13:33
like combining them may not so combining
00:13:36
them yes so to to the question of
00:13:38
combining them um initially we thought
00:13:41
that well perhaps we could try to
00:13:43
combine them we've done some preliminary
00:13:45
experiments in that way and in in some
00:13:48
way C6 may actually prevent yamanaka
00:13:51
factors from doing what they want to do
00:13:55
because C6 doesn't want to allow the
00:13:58
genome to completely
00:14:00
rewire um so in a way they could be
00:14:04
antagonistic to each other it can if if
00:14:07
let's say we wanted to um complete
00:14:10
reprogramming to take a cell and turn it
00:14:13
into embryonic stem cell so C six would
00:14:16
actually be an impediment to this
00:14:19
process because it wouldn't let the
00:14:21
whole epigeneium to open up and then
00:14:24
assume new structure um if we only want
00:14:27
to do it in partially so there may be
00:14:30
benefit in combining them so maybe C6
00:14:33
would not let the cell reprogram
00:14:35
completely which is not what we want in
00:14:38
the case of um somatic Rejuvenation but
00:14:42
maybe it will kind of limit uh so this
00:14:46
is something we actually want to study
00:14:50
in the future so that's one our
